Design of probes for species specificity of viruses belonging to morbillivirus

Abstract
Objective: To design and prepare the microarray probes for species specificities of measles virus, canine distemper virus, rinderpest virus, seal distemper virus, dolphin distemper virus and Peste des petits ruminants virus. Methods: All the sequences of genomes of above-mentioned viruses in GenBank were downloaded from ncbi web site and blasted by using biological software CLUSTAL W. The meaningful specific sequences of each virus were obtained and used as the candidate sequence for design of oligo probes by biological software Primer Premier 5.0. Gene chip was prepared with the optimal probes and verified for specificity. Results: Six oligo probes for species specificity, at even lengths with good specificities, were obtained, of which the temperatures for annealing were similar. Conclusion: Design of microarray probes for viruses by using GenBank and biological software Primer Premier 5.0 was rapid and effective.
